Single‐Molecule Force Measurement Guides the Design of Multivalent Ligands with Picomolar Affinity

An optimized picomolar‐affinity ligand was designed on the basis of the geometry of appropriate linkers for distinct multivalent binding modes. In their Communication (DOI: 10.1002/anie.201814347) Z. Li, S. Jiang, and co‐workers determined the actual distribution and separation of vascular endothelial growth factor receptors (VEGFR) on living cells by using a single‐molecule force measurement platform. The receptor distribution was then used to guide the rational design of a picomolar‐affinity tetravalent ligand and a positron emission tomography (PET) radiotracer.
