Optimal dosing of zoledronate to prevent fractures: improving risk‐benefit balance
AbstractIn their prospective, randomized, placebo‐controlled, double‐blind trial that investigated anti‐fracture efficacy of zoledronate in a total of 2000 osteopenic postmenopausal women aged over 65 years, Reid et al. [1, 2] provides crucial evidence that is highly useful for clinical practice; intravenous administration of zoledronate at a dose of 5 mg every 18 months for 6 years reduced fracture risk independently of baseline clinical characteristics including age, dietary calcium intake,...

Mass Spectrometry-Based Proteomics Analyses Using the OpenProt Database to Unveil Novel Proteins Translated from Non-Canonical Open Reading Frames
OpenProt is a freely accessible database that enforces a polycistronic model of eukaryotic genomes. Here, we present a protocol for the use of OpenProt databases when interrogating mass spectrometry datasets. Using OpenProt database for analysis of proteomic experiments allows for discovery of novel and previously undetectable proteins.

Ex Vivo Expansion of Hematopoietic Stem Cells from Human Umbilical Cord Blood-derived CD34+ Cells Using Valproic Acid
Here, we describe the ex vivo expansion of hematopoietic stem cells from CD34+ cells derived from umbilical cord blood treated with a combination of a cytokine cocktail and VPA. This method leads to a significant degree of ex vivo expansion of primitive HSCs for either clinical or laboratory applications.

Pupillary Response as Assessment of Effective Seizure Induction by Electroconvulsive Therapy
Pupillary responses (light reflex) were measured for assessment of adequate seizure induction by electroconvulsive therapy using an automated infrared pupillometer immediately after electrical stimulation. Constriction ratio was calculated and compared with seizure quality.

Interrogating Individual Autoreactive Germinal Centers by Photoactivation in a Mixed Chimeric Model of Autoimmunity
This protocol describes the generation of mixed murine bone marrow chimeras with spontaneous autoimmune germinal centers, in which autoreactive lymphocytes carry a photoactivatable green fluorescent protein (PA-GFP) reporter. This provides the ability to link cellular location in tissues with downstream molecular and functional analyses.

Genome Editing in Mammalian Cell Lines using CRISPR-Cas
CRISPR-Cas is a powerful technology to engineer the complex genomes of plants and animals. Here, we detail a protocol to efficiently edit the human genome using different Cas endonucleases. We highlight important considerations and design parameters to optimize editing efficiency.

Coelomic Fluid of Lumbricus rubellus Synergistically Enhances Cytotoxic Effect of 5-Fluorouracil through Modulation of Focal Adhesion Kinase and p21 in HT-29 Cancer Cell Line
Coelomic fluid of Lumbricus rubellus (CFL) has attracted interest due to its pharmacological properties, including antitumor effect. Furthermore, it is necessary to evaluate the response to treatment with new cancer therapeutic agents. This study aims to investigate whether the combination of CFL and 5-fluorouracil could reduce FAK protein level and iCa2+ and enhance p21 level. Furthermore, it is necessary to evaluate the response to treatment with new cancer therapeutic agents. After 24 hours of...

Variability in the organization and size of hunter-gatherer groups: Foragers do not live in small-scale societies
Publication date: June 2019Source: Journal of Human Evolution, Volume 131Author(s): Douglas W. Bird, Rebecca Bliege Bird, Brian F. Codding, David W. ZeanahAbstractMobile hunter-gatherers are often characterized as living in small communities where mobility and group size are products of the environmentally determined distribution of resources, and where social organization is multi-scalar: groups of co-residents are nested within small communities that are, in turn, nested within small-scale societies....

New bracketing luminescence ages constrain the Sima de los Huesos hominin fossils (Atapuerca, Spain) to MIS 12
Publication date: June 2019Source: Journal of Human Evolution, Volume 131Author(s): Martina Demuro, Lee J. Arnold, Arantza Aranburu, Nohemi Sala, Juan-Luis ArsuagaAbstractRecent chronological studies of the Sima de los Huesos (SH) hominin fossil site, Atapuerca, Spain, have established a close minimum age of at least 430 ka for sedimentary material immediately overlying the human remains. However, a firm maximum age limit still needs to be established for the SH fossils in order to better constrain...

Comparative Quantitative Study on Phytochemical Contents and Antioxidant Activities of Capsicum annuum L. and Capsicum frutescens L.
The variabilities in the phytochemical contents and antioxidant activities of four varieties of two closely related cultivated Capsicum species, C. annuum and C. frutescens, were examined as an additional tool for establishing their phylogenetic relatedness and for breeding purposes. The methodology involved the use of ethanolic and aqueous extractions for the determination of the phytochemical and antioxidant properties. The phytochemical contents including total flavonoid, total phenol, and proanthocyanidins...
